In this video, Michael Gass of PDS Equipment walks through the process of creating a gloss layer with a UV printer and Adobe Illustrator. To sum it up:

When creating a gloss layer, there needs to be two layers in the design.
Once the original layer is duplicated, do an image trace on the second layer.
Save the file for the gloss layer and the file for the image as two separate files.
